Solid Phase Extraction (SPE), also known as Solid Phase Extraction, is a widely used and popular sample pretreatment technique that uses solid adsorbents to adsorb target compounds from liquid samples, separate them from the sample matrix and interfering compounds, and then use eluent or heating desorption to achieve the goal of separating and enriching target compounds. It is developed on the basis of traditional liquid-liquid extraction (LLE) using the principle of similar phase solubility based on similar interactions between substances, combined with the basic knowledge of commonly used liquid chromatography (HPLC) and gas chromatography (GC) stationary phases.
In the solid-phase extraction process, the attraction of the solid phase to the analyte is greater than that of the sample mother liquor. When the sample passes through the SPE column, the analyte is adsorbed on the solid surface, and other components pass through the column with the sample mother liquor. Finally, they are eluted with an appropriate solvent. SPE can be divided into four types based on its principles: reverse SPE (RP-SPE), forward SPE (NP-SPE), ion exchange SPE (IE-SPE), and adsorption SPE. In most cases, solid-phase extraction is applied to process liquid samples for the extraction, concentration, and purification of volatile and semi volatile compounds in the samples; When used for solid samples, the solid sample needs to be processed into a liquid state first.
